Are polynucleotides worth the hype? The honest verdict on beauty's biggest buzz ingredient

Search trends don't usually impress me. But when an ingredient logs a 500% increase in search volume over a single year, I feel professionally obliged to pay attention. Polynucleotides — also sold under their scientific name PDRN — have gone from niche clinic treatment to full-blown beauty phenomenon in the space of about eighteen months. They are on the shelves at Boots. They are all over your skincare algorithm. And yes, they come from salmon DNA, which is exactly as interesting as it sounds.


So are they worth it? Here's what the evidence actually says.

What are polynucleotides, and what is PDRN?


Polynucleotides are long chains of DNA fragments. PDRN — polydeoxyribonucleotide — is the specific active compound used in most of the treatments you'll encounter: it's extracted from the sperm or roe of salmon, purified, and processed for use in both clinical and cosmetic settings. If that detail makes you hesitate, the Korean aesthetic medicine industry would like a word; they have been using it in injectable skin treatments for decades.


The reason it ended up in skincare is not aesthetic novelty. PDRN has a long clinical history in wound healing and post-surgical tissue repair. It works by activating adenosine A2A receptors in the skin — a pathway that triggers fibroblast proliferation, collagen and elastin synthesis, and an anti-inflammatory response. In plain terms: it signals to your skin's repair cells that there's work to be done, then gives them the building blocks — pre-made DNA nucleotides — to do it faster and with less metabolic effort. That's not a marketing claim. That mechanism has been confirmed in published research.


What the science actually shows


The evidence base for polynucleotides skincare has sharpened considerably in the last two years. A systematic review published in 2025, evaluating nine clinical studies across 219 patients, found significant improvements in skin elasticity (around 18.7%) and hydration (around 23.4%), alongside measurable reductions in wrinkle depth. A randomised, double-blind trial published in PLOS ONE in July 2026 compared a 0.1% PDRN eye cream against 0.1% retinol — and the PDRN formulation came out ahead on dermal thickness and crow's feet reduction over 28 days, with considerably less irritation.


Those are genuinely encouraging results. But there is a distinction that gets almost entirely lost in the social media coverage, and it matters a great deal.

Injectable vs. topical: they are not the same thing


Most of the robust clinical data — the RCTs, the Phase III trials, the long-term follow-up studies — relates to injectable PDRN treatments. Brands like Ameela, Plinest and Rejuran deposit the active compound directly into the dermis, bypassing the skin barrier entirely. The results from that delivery method are well-documented: tissue repair, collagen remodelling, improved skin quality over a course of sessions. The mechanism is not in question.


Topical polynucleotide serums face a more complicated picture. The issue is the 500 Dalton Rule: standard PDRN and polynucleotide fragments are far larger than the 500 Da threshold at which molecules can reliably penetrate intact human skin. Applying them in a serum, without any specialist delivery system, means they are most likely working at the surface — functioning as a humectant and film-former rather than triggering the deep cellular repair that makes injectable treatments effective.


This is not to say topical serums are useless. Emerging research using encapsulated formulations — lipid nanoparticles, exosomes, low-molecular-weight PDRN — suggests meaningful penetration is achievable, and results for these newer delivery formats are promising. Rejuran's topical range uses what they call c-PDRN, a smaller molecule developed specifically for better absorption. A 2025 plant-derived (peony) PDRN study showed real improvements in periorbital elasticity from topical application. And a comparative study from the Korean Dermatological Society found that a 1% topical PDRN formulation achieved approximately 78% of the wrinkle reduction seen with the injectable equivalent over twelve weeks — which is a meaningful result, even if it confirms the gap.


The honest read: topical serums with good formulation technology are not nothing. But they're not injectables, and the ingredient itself does not automatically make a serum effective. What matters is how the PDRN is delivered.


Realistic expectations and who it's actually for


For topical PDRN serums, think hydration, surface radiance, improved skin texture, and barrier support. These are real benefits; they're just not the same as tissue-level regeneration. If you use a well-formulated serum consistently, you should notice your skin looking more settled, plumper, and better-lit over eight to twelve weeks. If you're considering injectable PDRN treatments, the evidence is considerably stronger, though the context is different. This is a clinical procedure, not a skincare product. A course typically runs to two or four sessions spaced a few weeks apart; in the UK, expect to pay anywhere from £275 to £450 per session, with London pricing at the higher end. Under current UK regulations, polynucleotide injections sit in the Amber tier of the government's tiered model for cosmetic procedures — meaning they require oversight from a regulated healthcare professional. Face-to-face consultation is mandatory; remote prescribing is not permitted. If a clinic isn't following those rules, that is your cue to leave.


Regime does not review invasive procedures, so I'll leave the injectable conversation there. The distinction is worth understanding, though, because a lot of the excitement around this ingredient is fuelled by injectable results being transposed onto serum packaging.

UK PDRN products worth knowing about


If you want to try a topical polynucleotide serum, the field has expanded considerably. Medik8's Exo-PDRN Prismatic+ Serum (around £79, available at Cult Beauty) pairs PDRN with exosomes for improved delivery. Medicube's PDRN Pink Peptide Serum (£16 to £22 at Boots and Amazon UK) is an accessible entry point with a loyal following. Rejuran's Healer Turnover Ampoule uses their c-PDRN format at around £45 to £60. For those avoiding animal-derived ingredients, VT Cosmetics' PDRN 100 Essence uses plant-derived alternatives.


What to look for on the label: encapsulated PDRN, c-PDRN, low-molecular-weight PDRN, or any mention of a delivery system (liposomes, exosomes, nanoparticles). A serum that lists PDRN without any of these is most likely working at the surface only.


The verdict


Polynucleotides are not hype dressed up in science. The mechanism is real, the injectable evidence is solid, and the topical research is catching up faster than I expected. This is an ingredient worth taking seriously.


What is overstated: the assumption that any serum with PDRN on the label delivers the same results as a clinical treatment. The science behind delivery matters as much as the ingredient itself. A poorly formulated serum is still a poorly formulated serum, regardless of what's in it.


What is genuinely promising: well-formulated topical PDRN as a hydration and skin-quality ingredient, with real but realistic benefits over consistent use. And injectable PDRN, for those choosing to explore it in a clinical setting, has an increasingly strong evidence base behind it.

If the ingredient had arrived five years earlier, the evidence wouldn't have supported the attention. It has arrived now, and just about enough of the science has landed to justify it. That's a fairly unusual position for a trending skincare ingredient to be in.


Worth the hype? Cautiously, and with the right expectations: yes.
